    Abstract

    AIM: To investigate the effect of endotoxin on the expression of hepatocyte GUSB mRNA and serum β-glucuronidase (β-GD) activity in mice.

    METHODS: Twenty-three BALB/c mice were divided into two groups: control (injection of Saline without endotoxin) (n = 5) and LPS (n = 18). In the Lipopolysa cc haride (LPS) group, after injection of endotoxin, mice were put to death at 1/2, 2, 4, and 24 hours by breaking cervical vertebra. Serum β-glucuronidase activity was detected by an enzyme kinetics method; and the expression of GUSB mRNA in hepatocytes was detected by semi-quantity reverse transcription-polymerase chain reaction (RT-PCR).

    RESULTS: In the LPS 1/2, 2 and 4 h groups, the activity of β-GD levels (3.96 ± 0.88, 4.40 ± 0.55, 5.65 ± 0.58 Fishman Unit, respectively) was significantly higher than that of the controls (2 ......
