Abstract
AIM: To ascertain the effectiveness of GeneChip for screening hepatocirrhosis-related genes.
METHODS: Total RNA was isolated from tissues of two cases of hepatic cirrhosis and one normal liver by the TRIzol method, and then purified, reverse-transcribed to cDNA incorporating biotin-labeling probe, hybridized with GeneChip (covering 18 400 transcripts, representing 14 500 distinct genes). Picture signals of fluorescence in gene array were scanned and compared by computer analysis of differential expression of the genes in two tissues.
RESULTS: Same differentially expressed genes were 1424 (9.82%) and same up regulated and down regulated genes were 980 and 444 respectively, in two hepatic cirrhosis and tissue specimens one normal liver tissue specimen.
