    Nested PCR demonstrated: Hepatocellular carcinoma had HBV-DNA replication

    Abstract

    AIMS To study the HBV-DNA replication state in hepatocellular carcinoma(HCC) tissues and para-HCC tissues.

    METHODS Nested PCR were performed from laparotomy specimens of 32 cases; the para-HCC tissues of each case were also done. The 2 pairs of primers were designed just cutting across the 2 gaps of DR₁ and DR₂, so the target gene was cccDNA-replication template of HBV-DNA. HBsAg and HBcAg imunohistochemistry was also done in each case of HCC and para-HCC tissues.

    RESULTS 5 cases of HCC(grade Ⅰ-Ⅲ) tissues and 10 cases of para-HCC tissues were cccDNA positive. The HBsAg and HBcAg positivity rates were high in the cancer cells of cccDNA(+) HCC cases. ......
