    Effects of Kupffer cells on hepatocyte aging and its mechanism in rats

    Abstract

    AIM To study the effect of Kupffer cell (KC) on hepatocyte (HC) aging and its mechanism in rats.

    METHODS The effect of aging of KC on protein synthesis and mitochondrial metabolism status of rat HC of various ages (6, 12, 18 and 24 months of age, n=5 for each group) and its relations with the altered cytokines production of the aged KC were investigated with a HC-KC coculture system in vitro.

    RESULTS The protein synthesis and mitochondrial metabolism levels of 24-month-old HC cultured alone were significantly decreased, compared with these of 6-month-old HC. Coculture of HC of 6 or 24-month-old with 6-month-old KC resulted in an increased protein synthesis and mitochondr ial metabolism status and with 24-month-old KC in decreased ones. Addition of LPS (10ng/L) amplified the effect of KC on HC. The effect of aging on KC led to a significan tly higher level of TNF, IL-8 and NO excretion and a downregulated excretion of prostaglandin E₂(PGE₂). Stimulation of KC of each group with LPS elevated TNF and NO levels. The IL-8 levels of 6- and 12-month-old groups were significantly increased and these of 18- and 24-month-old groups statistically unchanged. The regularity of PGE₂ was just contrary to that of IL-8. ......
