Apoptosis in human hepatocellular carcinoma by terminal deoxynucleotidyl transferase-mediate

Abstract
AIM To examine the occurrence of apoptotic cell death in HCC-9204 cell line and 21 formalin fixed, paraffin-embedded biopsy specimens from liver cirrhosis (6 cases) and hepatocellular carcinoma (15 cases).
METHODS Apoptosis of HCC-9204 cell line was induced by adriamycin (20μmol/L). Modified TUNEL assay was conducted using in situ cell death detection kit fluorescein (Boehringer Mannheim). Sections were pretreated by microwave. Control experiments were also performed, including DNase-digestion before TUNEL as positive control and TdT and FITC-dUTP deletion as negative control. Routine HE staining was conducted on serial sections. ......
