肝星状细胞激活的内在机制

摘要激活的肝星状细胞是肝纤维化时产生细胞外基质的主要细胞.细胞因子的旁分泌和自分泌作用、氧化应激、微环境ECM的改变是激活肝星状细胞的主要外部机制,激活时最初的显著变化是细胞质内脂滴和维生素A减少甚至消失,并表达a-平滑肌肌动蛋白.脂滴(含中性脂肪)的存在意味着向脂肪细胞分化,肝星状细胞向脂肪细胞分化及去分化与其激活有何关联,是激活时重要的分子机制之一,还是仅仅为一种伴随现象?最近的工作表明,与脂肪细胞分化密切相关的C/EBP-a和PPARg基因表达下调及维生素A信号转导削弱是肝星状细胞激活的重要内在机制.如果能从肝星状细胞激活的外部和内在机制两方面都进行干预的话,有可能提高肝纤维化的疗效.
