脂多糖刺激对小鼠CDllc + 细胞MHC-Ⅱ类分子和B7-2分子表达的影响

【摘要】 目的 观察脂多糖刺激对小鼠CDllc + 细胞MHC-Ⅱ类分子和B7-2分子表达的影响。方法 用细胞因子定向诱导培养的方法在体外使小鼠骨髓细胞分化为树突状细胞(DC)。在形态学观察的基础上,采用流式细胞术检测细胞表面CDllc分子的表达,对小鼠DC及其纯度进行鉴定,并观察脂多糖刺激对小鼠CDllc + 细胞MHC-Ⅱ类分子(I-A b )和B7-2分子(CD86)表达的影响。结果 培养6d后平均每只小鼠可收获约(1.2~1.6)×10 7 个细胞,其中CDllc + 细胞为70.8%±6.3%。未经脂多糖刺激时,CDllc + I-A b+ 双阳性细胞为17.3%,CDllc + 、CD86 + 双阳性细胞为6.6%,占CDllc + 细胞的25.4%和10.0%;而经脂多糖刺激后,上述两类双阳性细胞分别增至50.7%和43.8%,占CDllc + 细胞的67.4%和63.8%结论 培养6d、未经脂多糖刺激的树突状细胞主要为未成熟型。关键词 树突状细胞 CDllc MCH-Ⅱ类分子 CD86 脂多糖
