黄芩对脂多糖致大鼠胶质细胞损伤的保护作用及机制

     【摘要】 目的 黄芩苷(Baicalin,Bai)是中药黄芩的主要有效成分之一,属于黄酮类化合物,有抑菌、抗炎、抗变态反应、抗氧化反应等药理作用,本文探讨黄芩苷对脂多糖所致星形胶质细胞(astrocytes,AC)损伤的保护作用及其机理。方法 体外培养新生SD大鼠原代AC,MTT分析法检测黄芩苷对正常组与脂多糖干预的星形胶质细胞细胞活力,逆转录多聚酶链式反应(RT-PCR)检测星形胶质细胞iNOS mRNA的表达。结果 5μmol,10μmol,20μmol浓度的黄芩苷能恢复LPS导致损伤的星形胶质细胞的活力(F=3.697,P=0.01)。脂多糖干预后iNOS mRNA的表达明显增多,黄芩苷可以抑制脂多糖所致的AC iNOS mRNA的表达(P<0.05,n=5),有量效关系。以20μmol的浓度黄芩苷对iNOS mRNA的表达抑制最明显。结论 黄芩苷能够抑制脂多糖导致AC的损伤,可能与其抑制iNOS mRNA的表达有关。
