人碱性成纤维细胞生长因子基因的克隆、表达及生物活性分析

【摘要】 目的 研究bFGF对缺血组织血管新生侧支循环形成的作用。方法 采用RT-PCR方法,从人扁桃体组织中扩增得到人bFGF cDNA,利用基因重组技术将该基因片段重组与pcDNA3.1(+)真核表达载体上,构建成pcDNA/b。应用脂质体介导的基因转移技术将重组质粒体外转染至人脐带静脉内皮细胞(HUVEC)。结果 重组质粒转染能明显促进内皮细胞的分裂增殖。进一步将重组质粒通过脂质体介导,体外转染至人肾细胞293细胞系进行表达,经G418筛选获得稳定表达的重组质粒细胞克隆。结论 表达产物经鸡胚绒毛尿囊膜试验表明有促血管生成活性。

【Abstract】 Objective To study the bFGF’s effect on the newly formed vascular bypass of ischemic tissue.Methods The human basic fibroblast growth factor(bFGF)cDNA was amplified by RT-PCR from human tonsil tissue,after DNA sequenced,the bFGF cDNAwas inserted into eukaryotic expression vector pcDNA3.1(+).The reˉcombinant plasmid pcDNA/b was transferred into HUVEC cells mediated by liposome.The transient expression of bFGF was detected by MTT,the results showed that bFGF protein was expressed in HUVEC cells72h after gene transfer.Results It had
