生血宁治疗失血性贫血及改善铁代谢相关基因的实验研究

【摘要】 目的 观察生血宁治疗失血性贫血的药效及对铁代谢平衡调节的基因机制。方法 将大鼠随机分成正常对照组、失血模型对照组、琥珀酸亚铁对照组100mg/(kg·d)、生血宁治疗组100mg/(kg·d),每组10只。除正常对照组外,其它3组每天尾端放血2ml,连续6天,使造成失血性贫血模型。第6天开始灌药或水。第6、16、24天取血测血常规,第24天取十二指肠标本用RT-PCR法测铁代谢相关基因DMT-1、IREG1、Hephaestin的mRNA表达量。结果 生血宁能提高红细胞计数、血红蛋白、红细胞比积、红细胞平均血红蛋白浓度、缩小红细胞体积分布宽度。生血宁在基因水平能改善铁代谢,降低失血性贫血大鼠铁代谢相关基因DMT-1、IREG1、ephaestin的表达,疗效与琥珀酸亚铁相似。结论 生血宁治疗失血性贫血效果明显;在基因水平能改善铁代谢,降低DMT-1、IREG1、Hephaestin。关键词 叶绿素盐 失血性贫血 铁代谢 阳离子转运蛋白 基因表达调节
