锝标记抗D-二聚体单克隆抗体在兔早期股动脉血栓定位诊断中的应用

【摘要】 目的 为评估锝标记抗D-二聚体单抗在兔早期股动脉血栓定位诊断中的应用。方法 将 99m Tc标记的抗D-二聚体单抗4D30注入兔体内,进行放射免疫显像。分别观察血栓部位和对侧相应部位的放射性分布,计算放射性比值;显像完毕后取血栓及周围肌肉组织,称重后测定单位质量的放射性计数及比值;取心、肝、脾、肺、肾及膀胱组织,观察病理学改变。结果 股动脉血栓形成6h内,注入显像剂后30min,血栓开始显像。血栓/血液和血栓/周围肌肉单位质量放射性比值分别为2.08±0.33,3.68±0.74。各器官均未见明显病理学改变。结论 提示 99m Tc-4D30可以用于实验性早期动脉血栓的定位诊断。关键词 锝 单克隆抗体 D-二聚体 血栓

【Abstract】 Objective To evaluate the uslefulness of technetium( 99m Tc)labelled monoclonal anti-D-dimer antibody(4D30)for target diagnosis of early femoral arterial thrombus of rabbit.Methods Radioimaging and quantitative radioactive analysis were performed by SPECT after 99m Tc-4D30was injected.Results Thrombus-to-blood and thrombus-to-muscle ratios were counted with gamma counter.The morphologic changes in heart,liver,spleen,lung,kidney and bladder were observed under light microscope.The femoral arterial thrombi showed at30min after injection(6h after the thrombi formed).Thrombus-to-blood and thrombus-to-muscle ratios were2.08±0.33and3.68±0.74.No pathologic change in all organs was observed under lightmicroscope.Conclusion It is concluded that the 99m Tc-laˉbelled anti-D-dimer monoclonal antibody(4D30)can be regarded as one of potential target agent in diagnosis of early femoral arterial thrombus. ......
