【摘要】 目的 探讨活性氧在血管紧张素Ⅱ诱导ECV304细胞增殖中的作用。 方法 体外培养人脐静脉内皮细胞株(ECV304),实验分为AngⅡ处理组、NAC干预组和正常对照组。采用改良MTT法、Fenton反应和硝酸酶还原法分别观察AngⅡ处理组、NAC干预组和正常对照组的ECV304细胞的增殖率和细胞产生ROS(·OH)的量。 结果 一定浓度的AngⅡ(0.03125~1μmol/L)作用12h时ECV304细胞增殖率增加,明显高于正常对照组(P<0.05);AngⅡ可诱导ECV304细胞产生ROS(·OH),ROS(·OH)的含量与ECV304细胞增殖率呈显著负相关;10mmol/L NAC可抑制1μmol/L AngⅡ对ECV304细胞的增殖作用,与正常对照组相比差异无显著性(P>0.05)。 结论 AngⅡ可诱导ECV304细胞产生ROS(·OH),ROS(·OH)含量与AngⅡ呈时间和剂量依赖性;NAC可抑制AngⅡ对ECV304细胞的增殖作用,这可能与减少ROS(·OH)的含量有关;ROS(·OH)可能是AngⅡ诱导的ECV304细胞增殖的主要信号转导分子之一。【关键词】 N-乙酰半胱氨酸 血管紧张素Ⅱ 活性氧 ECV304细胞 增殖

【Abstract】 Objective To explore the role of reactive oxygen species in the proliferation of ECV304induced by AngⅡ.Methods The lines of human umbilical vein endothelial cell(ECV304)cultured in vivo were divided into three groups which were treated by AngⅡ,AngⅡ+N-acetyl-L-cysteine(NAC),and normal culture medium.First we observed the proliferous effect of ECV304induced by AngⅡat different concentration with improved MTT and microscope.Then the contents of ROS(·OH)in three groups were detected by spectrophotometer.Results ECV304incubated with AngⅡ(0.03125~1μmol/L)for12hours increase the proliferation rate(P<0.05vs con-trol group);It is significant that the negative correlation between proliferation rate and the content of ROS(·OH);NAC(10mmol/L)can inhibit the proliferation of ECV304induced by AngⅡ(P>0.05vs control group).Conclusion ECV304induced by AngⅡcan produce ROS(·OH) ......
