【摘要】 目的 探讨葛根素在高浓度葡萄糖条件下对血管内皮细胞的内皮素转换酶-1b的影响。 方法 将人脐静脉内皮细胞株ECV304分别培养在含5.5mmol/L葡萄糖(对照组)、22mmol/L葡萄糖、22mmol/L葡萄糖+0.01g/L葛根素、22mmol/L葡萄糖+0.1g/L葛根素、22mmol/L葡萄糖+1g/L葛根素的培养基中培养24h,比色法测定细胞培养上清液的乳酸脱氢酶活力、丙二醛和一氧化氮的浓度,逆转录聚合酶链反应检测细胞中内皮素转换酶-1b的mRNA。 结果 各组乳酸脱氢酶活力和一氧化氮的浓度的差异无显著性,高浓度葡萄糖组中丙二醛和内皮素转换酶-1bmRNA比对照组增高,而含不同浓度葛根素组中的丙二醛浓度和内皮素转换酶-1b mRNA比22mmol/L葡萄糖组降低,其差异有显著性。 结论 葛根素可拮抗高浓度葡萄糖对内皮细胞的损伤,并抑制内皮素转换酶-1b mRNA的表达,此变化可能与改善血管收缩平衡有关。【关键词】 葛根素 葡萄糖 内皮素转换酶-1 内皮细胞
Effects of puerarin on expressions of engothelin-converting enzyme-1bmRNA induced by glucose in vascular endothelial cells

【Abstract】 Objective To investigate the effects of puerarin on expression of endothelin-converting enzyme-1bmRNA(ECE-1bmRNA)induced by high glucose in vascular endothelial cells(VEC).Methods Cultured hu-man umbilical vein endothelial cells(ECV304)were cultured with various concentration of glucose(Glu)and puerarin(Pue)(5.5mmol/L Glu、22.0mmol/L Glu、22.0mmol/L Glu+0.01g/L Pue、22.0mmol/L Glu+0.1g/L Pue、22.0mmol/LGlu+1g/L Pue)for24h.The release of lactate dehydrogenase(LDH)and maleic dialdehyde(MDA)con-tent in supernatant-conditioned media were measured by colori metric assay;The level of nitric oxide(NO)was e-valuated by nitrate reductase method;the expression of endothelin-converting enzyme-1b mRNA was determined by reverse transcription-polymerase chain reaction(RT-PCR).Results The releases of LDI and the level of NO were no differences in difference groups;The MDA contents and the expressions of ECE-1b mRNA in22.0mmol/L Glu were significant higher than that in control group;The MDA contents and the expressions of ECE-1b mRNA were significant decreased by puerarin treatment.Conclusion Puerarin has protective effect on injury and inhibit the expressions of ECE-1b mRNA on VEC induced by high glucose ......
