摘 要: 目的 观察脑源性神经营养因子(Brainderived-neurotrophic factor,BDNF)对帕金森病(Parkinson disease,PD)大鼠模型黑质多巴胺能神经元的影响。方法 将6-羟基多巴胺用立体定向法注入大鼠一侧中脑黑质制作PD大鼠模型,并在注入6-羟基多巴胺之间每天向预伤侧壳核区定位注入BDNF,采用酪氨酸羟化酶免疫组化方法及透射电镜等观察BDNF对黑质多巴胺能神经元的显微超微结构影响。结果 BDNF能改善6-羟基多巴胺造成的PD大鼠黑质多巴胺能神经元数目减少及超微结构影响。结论 BDNF可减轻6-羟基多巴胺对黑质多巴胺能神经元的毒性作用。

Abstract : Objective To study the effects of brainderived neurotrophic factor on substantial nigra dopaminergic neurons in Parkinson diseaserats. Methods Ratswere lesioned by unilateral injection of 6-OHDA into the substantial nigra in order tocreat animal models of Parkinson disease. Before injection of 6-OHDA,we infused BDNF intothe putaman for 2 weeks,then counted the numbers of TH positive neurons and observed themicroscopic,ultrastructure changes by tyrosine hydroxylase immunohistochemistry andtransmission electron microscope. Results In groups treated by BDNF,the numbers of dopaminergicneuron significantly increased in comprision to control groups. The microscopic andultrastructure also changed slightly. Conclusion BDNF can reduce and prevented dopamine neurons againstneurotoxin of 6-OHDA in vivo.
