摘要:目的 探讨地塞米松(Dex)在急性肝损伤中的作用及机制。方法 用D-半乳糖胺(D-GalN)加内毒素脂多糖(LPS)制成大鼠急性肝损伤模型,并用Dex预处理。结果 Dex组肝损伤明显减轻,伴有血中一氧化氮(NO)含量、肝组织中MDA及铁含量降低,SOD活性及锌含量增高。结论 Dex对此种肝损伤有保护作用,这种保护作用可能与其抑制NO过量产生,抑制脂质过氧化反应,增加组织抗氧化活性有关。

Antioxide function of dexamethasone and it′s protective effect on the acute liver injury induced by D-GalN/LPS

Abstract Objective To explore the effect of dexamethasone (Dex) on the acute liver injury induced by D-Galactosamine (D-GalN)/lipopolysaccharide(LPS) and the mechanism of its action.Methods Rats were injected intraperitoneally with D-GalN 600 mg/kg plus LPS 0.1 mg/kg and those in the Dex group were pretreated by Dex 4 mg/kg.Results The results showed that serum ALT (375±342 U/L),AST(683±367),TBil(3.21±2.83) in Dex group were significantly lower than that in model group (ALT,2352±698;AST,3548±285;TBil,20.71±5.92 P<0.01).Besides,Dex reduced the elevation of NO in serum and MDA,iron in liver tissues,while SOD and zinc changes were in opposition to MDA.Conclusion The results suggested that Dex has protective effects on the acute liver injury.This function may be correlatted with that Dex can inhibite lipid peroxide and increase the tissues antioxidation.
