摘要 目的:建立一种准确、快捷的P 53基因突变检测方法,为研究恶性肿瘤与抗癌基因P 53突变的相关性提供新的手段。方法:通过出芽酵母基因工程技术判断在酵母中P 53蛋白机能状态,以诊断P 53基因突变。结果:P 53机能诊断法检测结果经序列分析法验证完全准确。142株各种人体肿瘤培养细胞P 53存在状态,突变型占71%(101/142)、野生型占27%(38/142)、混合型占2%(3/142)。并证明非霍奇金氏淋巴瘤P 53存在状态与其化疗敏感性、生存期无明显相关性。结论:P 53机能诊断法可用作筛选大量的肿瘤培养细胞和临床肿瘤病理标本的P 53基因突变。方法简捷、诊断准确,是P 53存在状态较为理想的诊断方法之一。
FUNCTIONAL DIAGNOSIS OF P 53 AND ITS APPLICATION
IN DETERMINATING CARCINOMA P 53 GENE MUTATION

Abstract Objective:To establish a exact and speedy method for investigating P 53 gene mutation,and the relationship between carcinoma and antitumor gene P 53 mutation.Methods:P 53 gene mutation was diagnosed by using spout yeast gene technology to judge the functional status of P 53 in yeast.Results:The P 53 functional diagnosis was exactly re-established by using a serial testing method.The existing status of P 53 in 142 strains of various cultured human carcinoma cells was found,and of them 71% were mutated-type(101/142),27% were wild-type(38/142) and 2% were mixed-type(3/142).And the existing status of P 53 in non-Hodgkins lymphoma is not related to its chemotherapeutic sensitivity and existing stage.Conclusion:Functional diagnosis of P 53 is useful for a large scale sreening of cultured carcinoma cells and the clinical pathological examination of P 53 gene mutation in carcinogenisis tissue samples.This method is a relative ideal tool for diagnosing P 53 existing status with its simple,speedy and exact character.
