【摘要】 目的 以自发性高血压大鼠(SHR)为研究对象,观察血管紧张素Ⅱ(AngⅡ)受体AT1 阻滞剂缬沙坦的降压作用和对血液及组织AngⅡ浓度的影响,探讨左室重塑与心肌AngⅡ水平的关系。方法 将雄性15周龄SHR分两组(n=6),缬沙坦30mg. kg-1. d-1 ,对照组用等量蒸馏水灌胃。用高压液相放免法(HPLC-RIA)测定治疗4周前后SHR血液、肾脏、心脏和动脉壁中AngⅡ浓度。结果 治疗组血压由(205.0±10.7)mm Hg下降到(150.0±6.8)mm Hg(P<0.01),左室重量/体重由(264.3±17.1) mg/100 g下降到(208.2±13.9)mg/100 g(P<0.01),左室心肌和动脉壁的AngⅡ水平相应降低(P<0.01),但血液和肾脏中AngⅡ水平上升(P<0.01)。结论 缬沙坦抑制左室重塑与在AT1 受体水平上阻滞AngⅡ和降低心肌组织AngⅡ含量有关。
Effects of valsartan on the angiotensin Ⅱ levels and ventricular remodeling

【 Abstract 】 Objective To study the relationship between left ventricularremodeling and tissue angiotensin Ⅱ (Ang Ⅱ ) content based on the observation of effects of the specificantagonist of AT1 receptor valsartan on blood pressure and Ang Ⅱ levels in blood and tissues ofspontaneously hypertensive rats(SHR). Methods Valsartan (30 mg . kg-1 . . d-1 )and equal volume of distilled water were adminstered to 15-week-old male SHRs of theexperimental (n=6) and control groups (n=6) respevtive ly for 4 weeks by gavage. Highperformance liquid chromatography-based radioimmunoassays (HPLC-RIA) was used to measureAng Ⅱ levels inplasma, kidney, left ventricle, and artery. Results Four weeks ′ treatment with valsartan on SHR lowered the blood pressure from205.0±10.7 mm Hg to 150.0±6.8 mm Hg (1 mm Hg=0.133 kPa) (P<0.01)and lowered the ratioof left ventricular weight to body weight from 264.3±17.1 mg/100 g to 208.2±13.9 mg/100g(P<0.01). Ang Ⅱ levelswere lowered in left ventricular myocardium and arterial wall (P<0.01) but increased inplasma and kidney (P<0.01). Conclusion Valsartan can inhibit tissue remodeling in left ventricle throughAT1 receptor and low Ang Ⅱ levels.
