【摘要】 目的 研究性激素对脑损伤后星形胶质细胞(Ast)反应及胶质瘢痕形成的影响。方法 采用免疫组化、免疫荧光染色及流式细胞仪(FCM)计数等方法,观察切除性腺及补充性激素后脑穿刺损伤鼠伤灶周围胶原纤维酸性蛋白(GFAP)免疫组化染色阳性细胞的形态、数量与比例变化。结果 切除性腺鼠脑损伤后,伤灶周围Ast反应较单纯损伤鼠明显,表现为胞体肥大,突起增粗、延长,GFAP免疫组化染色显著增强。补充性激素后,Ast反应程度减弱,但未恢复到正常。切除性腺及用性激素治疗前后,Ast数量与比例无显著变化。结论 性激素能显著影响Ast形态变化,但对Ast的分裂、增殖作用不明显。

The effect of gonada hormone on reaction of astrocyte afterbrain stab injury

【Abstract】 Objective Tostudy the effect of gonadal hormone on reaction of astrocyte and glial scar formationafter brain stab injury (BSI). Methods The morphological changes andthe number of GFAP-staining positive astrocyte surrounding the regions of BSI were studiedby using immunohistochemistry, immunoinfluorescence stain and flow cytometry (FCM) after orchedectomy, overiectomy and gonadal hormone treatment. Results Thecastrated adult male or female rats resulted in more obvious morphological changes of GFAPpositive cells than those in intact adult rats after BSI. The cell body became more hypertrophic, the staining of GFAP more intense. FCM studies showed that the ratio ofastrocytes was not changed after BSI or treatment with gonadal hormone. Conclusion Thegonadal hormone can significantly cause the morphological changes of the astrocytes, buthas no marked effects on differentiation and proliferation of astrocytes after BSI.
