【摘要】 目的 研究β-磷酸三钙(β-TCP)和骨形态发生蛋白(BMP)的复合物(β-TCP/BMP)诱导成骨的作用。方法 在理化性质检测和生物安全性评价的基础上进行小鼠体内诱导成骨的实验,评价该材料的骨诱导能力。结果 多孔β-TCP钙磷比为1.5,平均孔径为400μm,平均气孔率为42.3%,抗压强度为0.826MPa(1MPa=7777.8mm Hg),在生理盐水中有一定的溶解度。溶血、急性毒性和致热源检测均在安全范围内。植入小鼠大腿肌袋72h有大量间充质细胞增生,1周可见软骨生长,4周有造血骨髓的板层骨出现,4周可见早期降解。结论 β-TCP/BMP是一种有骨诱导能力的人工材料。
Experimental study of ectopic bone induction of composites of porous β-tricalcium phosphate and bone morphogenetic proteins

【Abstract】 Objective To study the ectopic bone induction of composites of β-tricalcium phosphate (β-TCP) and bone morphogenetic proteins (BMP) as a kind of substitutes for bone graft. Methods The physic and chemical properties of the materials were detected and the biological safety was evaluated. The osteoinductive ability of the composites was measured in mice. Results The ratio of calcium to phosphate of the porous β-TCP was 1.5; the average diameter of the pores was 400 μm; the average porosity was 42.3%; and pressure strength was 0.826MPa(1 MPa=7777.8mmHg). The material was found to have some solubility in saline in vitro. The data of hemolysis, acute toxicity and source of fever were found within normal range. After the composites were implanted into the thighs of mice, the abundant mesenchymal cells were found at 72 h, cartilage at 1 week, lamellar bone with bone marrow at 4 and 8 weeks. No bone or cartilage was found in control group. The evidence of degradation of the materials was found at 4 weeks.Conclusion β-TCP/BMP composite is a kind of synthetic material with capacity of bone induction.
