【摘要】 目的 为探讨弥可保(甲钴酰胺)对糖尿病周围神经病变的治疗作用。方法 对链脲佐菌素(STZ)诱导的糖尿病周围神经病变大鼠早期给予弥可保500μg/kg肌肉注射,每天1次,2个月后行摇尾试验,测定坐骨神经运动传导速度和局部血流量,并分离腓肠神经,光镜下行腓肠神经形态学定量分析。结果 弥可保治疗后大鼠热痛阈值显著低于糖尿病组(P<0.01),但坐骨神经运动传导速度和血流量差异不显著(P>0.05)。大鼠腓肠神经有髓鞘神经纤维数量和密度显著高于糖尿病组,神经轴索和髓鞘显著大于糖尿病组(P<0.01)。结论 弥可保对实验性糖尿病鼠周围神经损害有一定的防治作用。

【Abstract】 Objective Tostudy the effect of methylcobalamin on peripheral neuropathy. Methods Streptozotocin-induced diabetic rats were given daily intramuscularinjection of methylcobalamin (500μg/kg) for twomonths. Threshold of heart-pain and motor nerve conduction velocity and circulation volumeof sciatic nerve as well as histomorphometric analysis were then compared between treatedand untreated diabetic rats and normal control rats.Results The threshold of heat-pain in the treated diabetic rats wassignificantly lower than that of the untreated diabetic group (P<0.01). But there was no significant difference of motor nerveconduction velocity and circulation volume of sciatic nerve between treated and untreateddiabetic groups. Morphometrical analysis showed that the number and density of myelinatednerve fibers, and the myclin sheath size and the axon size were significantly higher inthe methylcobalamin treated diabetic rats than in the untreated rats (P<0.001). Conclusion These findings suggest that methylcobalamin has some effect in theprevention and treatment of peripheral neuropathy in diabetic rats.
