【摘要】 目的 观察黄芪对糖尿病肾病(DN)的作用。方法 以STZ诱导糖尿病动物模型,采用免疫组化和RT-PCR检测方法观察黄芪对糖尿病大鼠肾皮质TGF-β蛋白、基因表达的影响。结果 糖尿病大鼠肾皮质TGF-β免疫染色强度比未治疗大鼠明显减轻,TGF-βmRNA水平比未治疗大鼠明显下降(P<0.01),黄芪与开搏通合用可增强上述作用。结论 黄芪可抑制糖尿病大鼠肾皮质TGF-β的过度表达,其防治DN发生、发展的作用机制可能部分是通过抑制TGF-β的过度表达。

    【Abstract】 Objective To observe the effectof Astragalus Membranaceus(AM) on diabetic nephropathy. Methods Theexpression of TGF-β protein and TGF-β mRNA in renal cortical samples of male STZ-induceddiabetic rats were determined by immunostaining and RT-PCR analysis. Results TGF-βimmunostain and TGF-β mRNA levels increased significantly in untreated diabetic rats ascompared with euglycemic ones (P<0.01). After eight weeks of AM treatment, the increased TGF-β immunostain and TGF-β mRNA levels declined significantly as compared withuntreated diabetic rats (P<0.01). These effects were enhanced if AM was combined with Captopril. Conclusion AM inhibits TGF-β protein and mRNAover-expression in renal cortex of diabetic rats suggesting that AM might play abeneficial role in experimental diabetic nephropathy, at least, partly by inhibiting TGF-βprotein and mRNA over-expression in kidney.
