摘 要:目的 研究VP-16诱导雄激素非依赖型前列腺癌细胞PC-3凋亡和抑癌基因p53表达的关系。方法 以末端标记法(TUNEL)和流式细胞术(FCM)研究PC-3细胞凋亡及细胞周期分布;以免疫组化方法研究p53蛋白的表达。结果 末端标记法显示凋亡的PC-3细胞呈明显的形态学改变,流式细胞术和免疫组化检测结果表明,随VP-16作用浓度的增加和作用时间的延长,PC-3细胞的凋亡率和其p53基因表达阳性的细胞百分率逐渐增高。结论 VP-16能够诱导PC-3细胞凋亡和增加p53基因表达,且具有作用浓度和作用时间依赖性。

Abstract:Objective To study the relationship between apoptosis induced by etoposide in androgen-independent PC-3 cells and to evaluate the expression of p53 tumor suppression gene.Methods TUNEL(TdT-mediated x-dUTP nick end labeling) and FCM (flow cytometry) were applied to investigate the apoptosis in PC-3 cells and distribution of cell cycle.p53 protein was detected immunohistochemically. Results Apparent apoptic morphological changes in PC-3 cells were observed by TUNEL.FCM and immunohistochemical assay indicated that the percentage of apoptic cells and the positive cells of p53 expression were increased with the increasing of exposure time and concentration of VP-16.Conclusions VP-16 can induce the programmed cell death of PC-3 cells and enhances the expression of p53 gene.
