【摘要】 目的 了解大鼠肾脏缺血及保存再灌注后E-selectin的表达情况。 方法 以E-selectincDNA为探针,应用Northern blot法分别测定缺血15、30分钟及保存24、48小时以及缺血、保存再灌注后8、16小时大鼠肾脏组织E-selectinmRNA的表达情况。 结果 实验显示缺血及保存的肾脏未见E-selectinmRNA表达,而缺血及保存再灌注后肾脏均见E-selectin mRNA表达,且保存肾脏在再灌注后早期E-selectinmRNA表达受到暂时性抑制。 结论 缺血及保存后再灌注是肾脏组织E-selectinmRNA表达的原因,保存再灌注后表达的暂时性抑制可能与低温有关。
E-selectin mRNAexpression in renal tissue suffered from ischemia and preservation-reperfusion injury

【Abstract】 Objective To study E-selectin expression in rat kidney suffered fromischemia and preservation/reperfusion injury. Methods Total RNA in rat renal tissues suffered from ischemia andpreservation/reperfusion injury was assayed by means of Chomzyski technique.The warmischemia time of the renal tissue has been 15、 30min,preserved for 24、 48h and reperfused for 8、 16h.Qualitative analysis of E-selectin mRNA was performedby Northern blot hybridization. Results E-selectin mRNA has been absent in ischemic and preserved kidneytissues but was expressed in all the samples after reperfusion.However,the expressionlevel of E-selectin mRNA has been temporally suppressed in early reperfusion. Conclusions Reperfusion induced the E-selectin mRNA in ischemic and preserved kidney tissues.Hypothermia or substrate in thepreservation solution might temporally suppress the E-selectin mRNA expression in theischemic and preserved kidney tissues in rats.
