【摘要 】 目的 探讨前列腺基质增生的机制。 方法 系用MTT和TUNEL法检测体外原代无血清培养的前列腺基质细胞的增殖，用免疫组化、RT-PCR方法检测平滑肌细胞特异性标记蛋白的表达。 结果 TGFβ有多重效应：浓度为0.01～10.00ng/ml的TGFβ可抑制指数增生期前列腺基质细胞增 殖；浓度低于0.1ng/ml的TGFβ刺激平顶期前列腺基质细胞增殖；浓度高于1ng/ml的TGFβ促增殖平顶期的成纤维细胞向平滑肌细胞分化。bFGF对于指数增生期和平顶期细胞均有刺激增殖的作用，并抑制增殖平顶期的成纤维细胞向平滑肌细胞分化。 结论 TGFβ₁ /bFGF比例的改变有调节前列腺基质细胞的稳态平衡的作用。

【Abstract 】 Objective Themechanism of prostatic stroma l proliferation was studied. Methods Cellcultures from th e prostatic stroma,including the fibroblasts and the smooth muscle cells (SMCs), had been established in serum free medium.The proliferation and apoptosis of thecell culture under the action of TGFβ₁ and bFGF was detected by MTT and TUNE Lmethods.The differentiation from fibroblasts to SMCs was deduced by measuring theexpression of SMC specific protein,myosin and smoothelin by the use of immunohistochemistry and RT-PCR methods. Results TGFβ₁ sh owedmultiple functions on the prostatic stromal cells according to both the wor kingconcentration and the growth period of cultures.(1)TGFβ₁ (0.01～10ng/ml)inhibited the growth of the prostatic stromal cells in exponent growth period;( 2)Lowerconcentration of TGFβ₁ (<0.1ng/ml) stimulated the growth of the conf luentcultures and had no influence on the differentiation,while higher concentr ation of TGFβ₁ (>1ng/ml) induced the differentiation and had little effect on the growth.bFGFstimulated the growth of the cultures and inhibited the differ entiation from fibroblaststo SMCs no matter what growth period they were in. Conclusions TGFβ₁ and bFGF may play important roles in regulating the stromal cell homeostasis.
