【摘要 】 目的 探讨线粒体DNA相对含量及基因缺失与衰老的关系。 方法 采用Southern杂交及聚合酶链反应(PCR)方法对48例不同年龄组人白细胞进行线粒体DNA缺失分析,用定量Southern杂交方法分析线粒体DNA含量。 结果 用Southern杂交方法未检测到线粒体DNA的缺失,用PCR方法检测到“普通缺失”21例,<40岁组20例中0例,40~49岁组4例中1例,50~59岁组2例中1例,60~69岁组10例中7例,>70岁组12例中12例。老年组线粒体DNA的相对含量明显高于其他组(P<0.01)。 结论 线粒体DNA缺失发生率及相对含量均随增龄而增加,为此,进行线粒体DNA缺失及含量的研究对衰老指标的测定及抗衰老措施的评估有意义。
Preliminary research of the relative content and deletion of mitochondrial DNA associated with aging

【Abstract 】 Objective To study the association of relative content and gene deletion of mitochondrial DNA (mtDNA) with aging. Methods The deletion of mtDNA of human white blood cells in 48 cases were detected by Southern blot and polymerase chain reaction (PCR). Furthermore the amount of mtDNA in these samples was analyzed by quantitative Southern blot . Results There was no detectable deletion of mtDNA in all the samples by Southern blot, however, “common deletion” could be detected in human white blood cells mtDNA in 21 of the 48 cases by PCR. The incidences of deleted mtDNA in different age groups are as follows: <40 years 0%; 40-49 25%; 50-59 50%; 60-69 70%; >70 100%. The amount of mtDNA in the elderly group is much higher than other two groups(P<0.01). Conclusions The proportion of deleted mtDNA to normal mtDNA is very low in normal tissue, but increases with aging. Only by PCR, the deleted mtDNA could be detected, while it is too difficult to be detected by Southern blot. The incidence of deleted mtDNA and the relative amount of mtDNA increase in elderly subjects and this change occurs after middle age.
