【摘要 】 目的 探讨腺伴随病毒(AAV)载体系统在纹状体有关结构中的转导特性。方法 以AAV-LacZ基因转导离体及在体纹状体神经细胞。用X-gal染色确定LacZ基因的表达。结果 AAV-LacZ基因在离体纹状体神经细胞的转导具有剂量与时间依存性特点;在在体纹状体神经细胞内的表达则可稳定持续至少长达3个月以上。结论 AAV载体系统可在纹状体有效转导并稳定表达外来基因。

【 Abstract 】 Objective Explore the transduction behavior of adeno-associated virus (AAV) vector within the structure of striatum. Methods Rat striatal nerve cells were transduced with AAV-LacZ in vitro and in vivo and β -galactosidase expression was assayed by using X-gal staining. Results In vitro β -galactosidase expression showed a dose and time-dependence and in vivo expression in the rat striatum maintained at least for three months. Conclusion AAV-mediated gene transfer in striatal nerve cells is efficient and stable.
