【摘要】 目的 研究FHIT基因在喉癌发生中的作用。方法 应用RT-PCR技术筛查了喉癌细胞系Hep-2、宫颈癌细胞系HeLa和20例原发性喉癌的FHIT基因转录本,对其中的2例异常转录本进行了测序分析。同时,对另外60例原发性喉癌FHIT基因进行了微卫星多态分析。结果 在20例喉癌中,14例(70.0%)显示FHIT基因异常转录本,Hep-2和HeLa细胞系FHIT基因转录本均存在缺失。测序发现,异常的FHIT基因转录本常缺失多个外显子,而且多发生于第5和(或)第8外显子。微卫星多态分析表明,FHIT基因在喉癌中既存在杂合性丢失,又存在微卫星不稳定现象,但以前者为主。结论 FHIT基因参与了喉癌的发生,可能是喉癌候选抑癌基因之一。

【Abstract】 Objective Todetermine the role of FHIT gene in laryngeal carcinogenesis. Methods Laryngealcancer cell line Hep-2, HeLa cell line and 80 primary tumors from patients with laryngealcancer were studied by means of RT-PCR, sequencing and microsatellite analysis. Results Abnormaltranscripts of FHIT gene were found in the two cell lines and 70%(14/20) of primarylaryngeal carcinomas. Aberrant laryngeal tumor transcripts often lack more than two exonsof FHIT gene. Microsatellite analysis showed that both loss of heterozygosity andmicrosatellite instability of FHIT gene existed in laryngeal carcinoma and the former wasmore common. Conclusion FHIT gene participates in the laryngealcarcinogenesis and may be one of the candidate tumor suppressor genes.
