【摘要】 目的 探讨微载体培养人肝细胞用于人工肝的可能性。 方法 用微载体Cytodex 3培养人肝细胞,将其置于中空纤维型生物反应器构成人工肝装置,在体外循环实验中进行生物功能及细胞活力的观察与评价。 结果 体外两步法分离的人肝细胞,在辅助促聚条件下有效地粘附于微载体上,呈悬浮生长状。移入生物反应器内进行体外循环实验6小时,见循环液中白蛋白含量明显增加,利多卡因浓度显著减少,实验结束时肝细胞仍保持良好活力。 结论 微载体培养人肝细胞为材料的人工肝,具备生物合成与转化代谢功能,可望对肝衰竭患者起支持与治疗作用。

【Abstract】 Objective To studythe possibility of using microcarriers cultural human hepatocytes for artificial liver. Methods Thebiologic function of artificial liver device, consisting of microcarriers cultural humanhepatocytes and a hollow fiber bioreactor was observed under circulating in vitro. Results Onthe conditions of facilitating aggreate, the human hepatocytes isolated by extracorporealtwo-stage method were successfully attached to cytodex 3 microcarriers, which grew assuspending in media, setting the hepatocytes into the bioreactor and circulating the mediafor 6 hours, a markedly increase of ablumin and decrease of Lidocaine were found. A goodviability of hepatocytes was also observed after the experiment. Conclusion Theartificial liver with microcarriers cultural human hepatocytes has both the biosynthesisand biotransformation function. It could be expected to support and treat the patients ofhepatic failure.
