【摘要】目的 观察庆大霉素对兔成骨细胞增殖及超微结构的影响。方法 从出生2周的新西兰兔颅盖骨获取成骨细胞,在含不同浓度庆大霉素的DMEM培养液中培养24小时或72小时,采用四唑盐比色法观察庆大霉素对成骨细胞增殖的影响,并对在含不同浓度庆大霉素的DMEM培养液中培养96小时的成骨细胞进行透射电镜观察。结果 高浓度庆大霉素组(800μg/ml、1600μg/ml)光吸收值明显低于对照组(P<0.01),高浓度庆大霉素组超微结构表现为内质网扩张,胞浆内见大量的吞噬溶酶体。结论 高浓度庆大霉素明显抑制成骨细胞增殖,呈现一定的毒性作用。
Effects of Gentamycin onProliferation and Ultrastructure of Primary Cultured Rabbit Osteoblasts

【 Abstract 】 Objective Toobserve the effects of gentamycin on the proliferation and ultrastructure of primarycultured rabbit osteoblasts. Methods Osteoblasts isolated from the two- week newly born rabbits werecultured for 24 h or 72 h in DMEM media with different concentrations of gentamycin. MTTmethod was used to observe the effect of gentamycin on cell proliferation. Theultrastructural changes of cultured osteoblasts exposed to gentamycin for 96 h were alsoobserved under transmission electron microscope. Results The opticaldensity values of higher concentration gentamycin groups (800μ g/ml, 1 600μ g/ml) were much smaller than that of the control group(P<0.01). The swelling of mitochondria and the presence of abundant phagolysosomes werecharacterized as the ultrastructural features. Conclusion Highconcentration gentamycin demonstrated a certain toxic inhibitive effects to osteoblasts.
