【摘要】 目的 解决修复重建组织器官缺损所需移植材料的问题,利用组织工程技术,进行组织工程牛透明软骨的实验研究。方法 用分离获得的牛软骨细胞,与医用组织引导再生胶原膜体外联合培养1周后,以软骨细胞可降解材料复合物的形式植入裸鼠体内,8周后对所获得的组织工程软骨样组织进行形态学研究。结果 在实验获得的组织工程软骨样组织中有软骨细胞的存在,并且可以分泌硫酸软骨素。证实为组织工程牛透明软骨。结论 实验结果表明应用组织工程技术,在体外进行组织工程软骨的制作,可能成为解决整形外科领域修复缺损用移植材料不足的方法之一。
A study on guided tissueregeneration and tissue engineering-transplantation of collagen membrane seeded withcultured hyaline chondrocytes

【 Abstract 】 Objective Lackof transplant material is a main problem in the repair and reconstruction of tissue ororgan defects. To seek the succedaneum of transplants, we studied and usedtissue-engineered growth hyaline cartilage.Methods The articular hyalinecartilage obtained from newborn calf within 6 hours of sacrifice was enzymaticallydissolved. The chondrocytes were then seeded onto the medical collagen membrane of guidedtissue regeneration in a 24 well plate in RPMI1640 medium. One week later, the chondrocytemedical collagen membrane of guided tissue regeneration complex was implantedsubcutaneously into the back of nude mice. Eight weeks after implantation, the animalswere sacrificed.Results The present study seems to demonstrate that thecartilage-like tissue was strong enough to be transferred after being implanted for 8weeks. The cartilage-like tissue was proved to be tissue-engineered hyaline cartilage byHE stain. The chondrocytes could secrete chondroitin sulfate as proved by Lev-Spicerstain. Conclusion Tissue engineering technique was used to maketissue-engineered cartilage in vitro. It will provide a new transplant material for therepair of tissue or organ defects.
