摘要 为观察灌注压力对肝移植物活性的影响,实验选择SD大鼠,经门静脉在不同压力下灌注4℃的Collin's液以保存肝脏,然后应用高效液相色谱仪检测各组的高能磷酸化合物,制备出肝细胞悬液后采用阳离子测定仪测定肝细胞内游离钙浓度,并同时观察了肝脏的超微结构变化及光镜病理改变。结果发现:随灌注压力的升高,肝脏的能量代谢、肝细胞内游离钙浓度以及超微结构均有明显改变。本实验结果提示:不适当的灌注压力可导致肝脏移植物的能量代谢的变化及肝细胞的钙超载,从而影响肝移植物的活力;为了取得高质量的供肝,应采用低温低压灌注。

    

THE PRESSURE OF INFUSION AFFECTING ON HEPATICTRANSPLANT

Abstract SD mice were selected forCollin's solution (4℃) infusion into the portal vein with different pressure to preservethe liver transplants. The following parameters were determined ①liver tissue aderineribonucleotide including adenosine triphosphate (ATP), adenosine diphosphate (ADP),adenosine monophosphate (AMP), ②cytoplasmic free Ca2+ in single liver cell([Ca2+ ]i ) and ③tissue pathologic ultra-structure change byhigh-performance liquid chromatography into quantimeter and pathologic examinationrespectively. The result suggested that with the infusion pressure becoming higher, theliver free Ca2+ ([Ca2+ ]i ), tissue aderine ribonucleotide, EC and tissue pathologic ultra-structure changed obviously. This resultshows [Ca2+ ]i , EC and tissue aderine ribonucleotide mightindicate the viability of liver transplant, and using low pressure infusion has benefiteffect on liver preservation.
