肠球菌随机扩增多态性DNA分型及应用研究

【摘要】 目的 建立随机扩增多态性DNA(random amplified polymorphic DNA,RAPD)分型技术,对肠球菌进行基因分型,探讨RAPD在分子流行病学中的应用,并探讨肠球菌的基因分型及其与耐药性的关系。 方法 对医院临床标本中分离的64株肠球菌进行RAPD基因分型,并分析基因型与耐药性的关系。 结果 RAPD分型结果显示,经优化的RAPD分型技术能有效地对肠球菌进行基因分型,分型率为100%。64株肠球菌根据扩增片段的差异分为8种基因型。以Ⅰ、Ⅱ、Ⅲ、Ⅶ型为主,共占81.4%。Ⅰ、Ⅷ型具有对β内酰胺类抗生素和高水平氨基糖苷类抗生素耐药。Ⅳ型具有对β-内酰胺类抗生素耐药。Ⅲ、Ⅶ具有对高水平氨基糖苷类抗生素耐药。Ⅱ、Ⅴ、Ⅵ型具有对β-内酰胺类抗生素、高水平氨基糖苷类抗生素、糖肽类抗生素敏感。 结论 RAPD分型技术是一种特异、敏感、简便快速、分辨力强、分型率高、重复性好的分型方法。可有效地对肠球菌进行基因分型,并且肠球菌的RAPD分型与其耐药性有一定关系。【关键词】 肠球菌;随机扩增多态性DNA;基因分型

【Abstract】 Objective To establish the random amplified polymorphic DNA(RAPD)genotyping,to discuss its role in molecular epidemiology.And to investigate the correlation of genotypes of enterococcus and its phenotype of drug re-sistance.Methods Using RAPD technique to genotype from clinical isolated samples from four hospitals and to analy-sis the relationship both the phenotype and drug resistance to antibiotics.Results Results of RAPD genotyping showed that RAPD in optimized condition could be used in enterococcus genotyping with the genotyping rate was100%.32strains of enterococcus were grouped into eight genotypes based on the fragments of amplified products.The most common types wereⅠ ......
