【摘要】 目的 T细胞介导的宿主抗移植物反应(host-versus-graft reaction,HVGR)是移植排斥的主要反应之一。本研究利用T细胞接种诱导特异性的免疫耐受来抑制同种心肌移植的HVGR。 方法与结果 (1)Con A(刀豆蛋白A)和抗CD 3 单克隆抗体能刺激同种抗原免疫小鼠淋巴细胞增殖,所制备的T细胞疫苗可显著延长移植心肌的存活期(P<0.05)。(2)接种T细胞疫苗后,体外实验证明机体可产生抗T细胞个体基因型细胞,调节受体的免疫系统对同种移植心肌产生特异性免疫耐受。(3)病理切片观察T细胞疫苗接种组移植心肌部淋巴细胞浸润明显减少。 结论 本实验研究初步证实T细胞疫苗可诱导机体产生对移植物的特异性免疫耐受,显著延长移植物的存活期,是一种新的有前景的抗移植排斥反应疗法。【关键词】 同种心肌;移植排斥;T细胞疫苗

【Abstract】 Objective It is one of the most important host-versus-graft reaction(HVGR)of T cell-mediated immunity in the rejection of transplantation.For inhibit HVGR of allomycardium transplantation,we tried to induce specific immune to tolerance byT cell vaccination.Methods and Results (1)The vaccines were prepared by activat-ing alloantigen-immunised mice T cell with concanvalin A(Con A)or CD 3 McAb could dramatically prolong the sur-vival of transplanted allomyocardium(P<0.01).(2)After T cell vaccination,it was confirmed in vitro that anti-idio-type cells were induced and they have the ability to regulated immunal network to achieve specific tolerance to specific allograft myocardium.(3)Histopathological examination indicated that infiltration lymphocytes of decreased obviously in transplanted allomycardium after T cell vaccination.Conclusion The results of the studies preliminary demon-strate:T cell vaccination enable to allomycardium of transplantation to survive longer and calculated it would provide a novel way for inhibiting transplantation rejection. ......
