Abstract
AIM: To explore the effects of olsalazine sodium on the expressionof nitric oxide synthase (NOS) in patients with ulcerative colitis (UC).
METHODS: The colonic mucosa specimens were collected from 36patients with active ulcerative colitis. The expression of inducible NOS(iNOS), epithelial NOS(eNOS), and nitrotyrosine were detected byimmunohistochemistry before and after the patients were treated witholsalazine sodium.
RESULTS: eNOS was expressed in the mucosal epithelial cells ofthe inflammatory area in 4 patients, but not expressed in the inflammatorycells. iNOS was expressed in the mucosal epithelial cells of all the 36(100%) samples (13 strong positive and 23 light positive) taken frominflammatory area. iNOS light positive staining in acinar cells of colonicepithelium was detected in 1 patient only. Inflammatory cells in thelamina propria expressed iNOS in 18 (50%) cases. After thetreatment of olsalazine sodium, iNOS was lightly and positively expressedin the mucosal epithelial cells of only 5 patients, and no iNOS wasexpressed in non-inflammatory areas. Nitrotyrosine was detected in themucosal epithelial cells of inflammatory areas in 5 patients, of whichonly 1 was accompanied by light and positive expression of nitrotyrosinein the acinar epithelial cells. No nitrotyrosine expression was found inthe inflammatory cells of the lamina propria and non-inflammatory areas.
CONCLUSION: Olsalazine sodium can inhibit the expression ofiNOS, alleviate the inflammation, and promote the healing of the ulcer.
