【摘要】 目的 观察丝裂素活化蛋白激酶(mitogen-activated protein kinase,MAPK)在神经肽Y(neuropeptide Y,NPY)及去甲肾上腺素(NE)促心肌细胞肥大过程中的作用。 方法 进行原代乳鼠心肌细胞培养,使用髓磷脂碱性蛋白为底物,γ- 32 P-ATP为磷酸基来源,测定在NPY、NE作用过程中MAPK活性的变化。 结果 NPY、NE作用后5min内,MAPK活性即有明显升高,在15min达高峰,25min后仍显著高于对照组(P<0.01);二者合用可使MAPK活性进一步增高(P<0.01)。 结论 作为一种快速、有力的信息传递工具,信息共同通路MAPK在NPY、NE生物学信息传导过程发挥了重要作用。【关键词】 丝裂素活化蛋白激酶(mitogen-activated protein kinase,MAPK);心肌细胞肥大;神经肽Y(NPY);去甲肾上腺素(NE)
【Abstract】 Objective To investigate the role of MAPK on the hypertrophic effects of cardiomyocytes induced by NPY and NE.Methods Neonatal Wistar cardio-myocytes were cultured first,then the role of MAPK on the hyper-trophic effects of cardiomyocytes were investigated through measuring the activity of plasma MAPK in different condi-tions.Results 1.The plasma MAPK activity was significantly ascended when NPY or NE was added(P<0.01vs Control);2.The plasma MAPK activity of NPY+NE group was significantly higher than NPY and NE group(P<0.01).Conclusion 1.The growth-promoting effects of NPY and NE were acted through MAPK signal route;2.Be-ing factors which were co-secreted from sympathetic nerves ......
