FHIT基因异常转录与宫颈癌相关性的研究

【摘要】 目的 探讨脆性组氨酸三联体(FHIT)基因与宫颈癌发病的关系。方法 采用巢式逆转录聚合酶链反应(nested RT-PCR)及DNA直接测序法对15例正常宫颈组织、30例宫颈癌组织及配对癌旁正常组织、宫颈癌Hela细胞系进行FHIT基因异常转录分析,并选择1例正常转录本及2例异常转录本进行DNA序列分析。结果 除7例宫颈癌组织及1例癌旁正常组织外,其余标本均扩增出FHIT基因野生型转录本。Hela细胞系同时存在野生型及异常转录本。FHIT基因的异常转录率在宫颈癌组织为47%(14/30),高于癌旁正常组织20%(6/30)及正常宫颈组织13%(2/15),差异均有显著性(P<0.05)。FHIT基因的异常转录与癌组织的生物学特性无关。DNA测序证明异常转录本为FHIT基因多个外显子的缺失及DNA序列的插入。结论 FHIT基因的异常转录是宫颈癌组织及Hela细胞系中的常见事件,可能与宫颈癌的发生有关。【关键词】 子宫颈肿瘤;基因脆性组氨酸三联体;逆转录聚合酶链反应;转录;遗传
Study on association of aberrant fragile histidine triad gene transcripts with human cervical cancer

【Abstract】 Objective To investigate the relationship between the fragile histidine triad (FHIT) gene and human cervical cancer.Methods Normal cervical tissues from 15 patients,cancerous and matched adjacent normal tissues from 30 patients with cervical cancer and Hela cell line were examined for abnormalities of the FHIT gene transcripts by nested RT-PCR.1 normal and 2 abnormal samples were selected for DNA direct sequencing.Results Wild-type FHIT gene transcripts were found in all samples except 7 cancerous tissues and 1 adjacent normal tissues.There were wild-type and aberrant transcripts in Hela cell line.Abnormal FHIT gene transcripts were obtained from 47%(14/30) cancerous tissues,20%(6/30) matched adjacent normal tissues and 13%(2/15) normal tissues.The rate of aberrant FHIT transcripts in cancerous tissues was significantly higher than that in their matched adjacent normal tissues and normal tissues(P<0.05).Clinicopathological analysis of patients showed that aberrant transcripts of the FHIT gene had no correlation with biological features of the tumors.Sequence analysis of the aberrant transcripts confirmed the absence of several exons of the FHIT gene and insertions of DNA sequences.Conclusion The expression of aberrant FHIT gene transcripts is common in cervical cancer and Hela cell line.Abnormalities of the FHIT gene may be associated with cervical tumorigenesis. ......
