消瘢方预防猪血清免疫性肝纤维化的实验研究

摘要:目的:探讨消瘢方对猪血清免疫性肝纤维化的预防作用。方法:60只SD雄性大鼠被随机分为4组:正常对照组、模型组、消瘢方高、低剂量预防组。除正常对照组外,所有大鼠均腹腔注射猪血清(1次/3 d,共12周),实验结束后处死大鼠,收取血清及肝组织标本,测定谷丙转氨酶(ALT)和谷草转氨酶(AST)、血清Ⅲ型前胶原(PCⅢ)及透明质酸(HA),利用HE及Masson染色对肝组织纤维化程度进行评价,并观察肝组织胶原的分布及α平滑肌肌动蛋白(α-smooth muscle actin, α-SMA)的表达。结果:消瘢方能显著降低肝纤维化大鼠血清ALT、AST、PCⅢ及HA水平,减少肝组织胶原的分布及α-SMA的表达,能显著改善肝纤维化程度。结论:消瘢方能有效预防肝纤维化的发生、发展,其作用可能与保护肝细胞,抑制细胞外基质合成及HSC活化有关。关键词:消瘢方;肝纤维化
The Preventive Effects of the XiaoBan Recipe on the Hepatic Fibrosis Induced by Porcine Serum

Abstract:Objective:To investigate the preventive effects of XiaoBan Recipe(XBR)on the hepatic fibrosis induced by porcine serum.Methods:Sixty male SD rats were randomly divided into four groups: normal control group, model group, low-dose-prevented group and high-dose-prevented group. Except the normal control group, the other rats were given an intraperitoneal injection of porcine serum(every three days for 12 weeks).At the same time, the two XBR-prevented groups were dealed with XBR, in the end of the experiment, the serum and livers of all the rats were sacrificd to detect alanine transaminase(ALT),aspartatc aminotransferase(AST), pre-collagenⅢ(PC Ⅲ), hyaluronic acid(HA).Then the degree of hepatic fibrosis was evaluated by HE,Masson stain. The distribution and content of hepatic collagen and α-SMA was observed.Results:XBR could obviously decrease the concentration of serum ALT, AST, PCⅢ and HA of rats with hepatic fibrosis, and reduce the stage of hepatic fibrosis. The distribution and expression of collagen and α-SMA were obviously less in prevented group than that in models group.Conclusion:XBR can prevent the occurrence and development of hepatic fibrosis in rats, the effect may be related to its preventing liver cell against injury, inhibiting the synthesis of extracellular matrix(ECM) and the activation of hepatic stellate cell(HSC). ......
