    摘要:目的 建立高效液相色谱法测定小鼠血浆及组织样品中氟尿嘧啶(5-Fu)的浓度,并进行5-Fu在小鼠体内的药物动力学研究。方法 以5-溴尿嘧啶(5-Bru)为内标,血浆和组织样品经液-液萃取后,采用SHIMADZU高效液相色谱仪进行分析,色谱柱为Kromasil C18 (4.6 mm×200 mm, 5 μm),流动相为乙腈-水 (1︰99,V︰V),检测波长为265 nm。结果 该法测定5-Fu在血浆及各组织中的平均回收率较高,日间、日内精密度符合有关规定。结论 方法操作简便、快速、适用于5-Fu在小鼠血浆、组织中的检测及药物动力学研究。

    Determination of 5-Fu in mice plasma and tissues by high performance liquid chromatography and pharmacokinetic study

    Abstract: Objective To develop a HPLC method for determining the concentration of 5-Fu in mice plasma and tissues, and study its pharmacokinetics. Methods 5-Bru was used as the internal standard and plasma and tissues samples were prepared based on a simple liquid-liquid extraction. The analytical column was Kromasil C18 (4.6 mm×200 mm, 5 μm). The mobile phase was acetonitrile- water (1∶99, V∶V). The UV detective wavelength was 265 nm. Results The extraction recovery of 5-Fu in plasma and tissues was high, and the precision of inter-day and intra-day accorded with relevant regulation. Conclusion It appeared to be an accurate, convenient, and effective method, and is suitable for the detection of 5-Fu in mice plasma and tissues.
