碱性成纤维细胞生长因子在实验性硬脑膜重建中的应用

     【摘要】 目的 探讨碱性成纤维细胞生长因子(bFGF)在实验性自体筋膜硬脑膜重建中的应用价值。方法 采用大鼠自体筋膜硬脑膜重建动物模型,通过免疫组织化学方法观察自体筋膜硬脑膜移植的愈合过程及细胞因子bFGF在其中的表达;并通过脑脊液漏研究、组织学及免疫组织化学方法研究外源性bFGF对自体筋膜重建硬脑膜愈合过程的影响及用RT-PCR方法研究内源性bFGF mRNA的表达情况。结果 大鼠自体筋膜移植硬脑膜部位bFGF表达明显;外源性bFGF应用1周后,其重建硬脑膜抵抗脑脊液漏压力、移植筋膜表面纤维层厚度及Ⅰ型胶原纤维表达均好于对照组;bFGF mRNA的表达没有明显变化。结论 在自体筋膜移植硬脑膜的愈合过程中,bFGF可能起了重要的作用;外源性bFGF处理的大鼠其移植筋膜愈合优于对照组。

    The application of basic fibroblast growth factor on experimental dural reconstruction achieved using a free fascial graft

    【Abstract】 Objective To study the effect of basic fibroblast growth factor on the experimental dural reconstruction achieved using a free fascial graft.Methods A dural reconstruction model was created in rats. The healing process and the effect of bFGF on it were studied by immunohistochemical method. The influence of exogenous bFGF on it was studied by CSF leak experiment, histological and immunological methods.Results The reconstructed dura was immunohistochemical stained with antibodies against bFGF. The pressures against CSF leak, the thickness of fibrous tissues around the fascia and the expression of type-1 collgen fiber in the dura 1 week after reconstruction with exogenous bFGF were better than those in control groups. There is no statistical difference in the expression of bFGF mRNA between the exogenous bFGF group and control groups.Conclusion bFGF may play an important role in the healing process of free fascial graft after reconstruction and exogenous bFGF could potentiate the process. ......
