和田地区维吾尔族长寿与非长寿家族DNA断裂水平研究

摘要: 目的: 研究和田地区洛浦县长寿与非长寿家族的DNA断裂水平的基础值及其相关因素。方法: 在征得调查对象同意后采集血液样本,用彗星实验方法检测其血淋巴细胞DNA断裂水平。统计分析采用t检验、秩和检验和相关分析等方法。结果: 长寿与非长寿家族的DNA断裂率及其校正均数差异都有统计学意义(P<0.01),断裂分级之间差异也有统计学意义(P<0.01),且断裂率与年龄呈正相关,有随年龄增高而增加的趋势,20~39岁组、40~59岁组及60~79岁组之间差异有统计学意义,但是性别之间比较差异无统计学意义(P>0.05)。结论:长寿与非长寿家族的DNA断裂率均数的基础值在排除年龄、性别、吸烟、饮酒等因素的影响后差异仍有统计学意义,长寿与非长寿家族寿命的长短可能与遗传背景产生的DNA损伤不同有一定的关系。关键词: 长寿家族; 非长寿家族; DNA断裂; 彗星试验
The study of longevity and nonlongevity families lymphocytes DNA strand breaks in HeTianSUN

Abstract: Objective: To determine the background levels of DNA strand breaks level and the correlation factors in longevity families and nonlongevity families people in the city LuoPu. Methods: Collect blood samples after getting agreement of participants .In laboratory we use comet assay to evaluate DNA damage in lymphocytes of these people. One hundred cells of each donor were graded by eye into 0~Ⅳ categories according to the amount of DNA in the tail. Results: The results of this study demonstratd the DNA damage frequency and the modify DNA damage rate of longevity families and nonlongevity families had statistically significance (P<0.01).The DNA damage frequency of different age groups: 20~39, 40~59, 60~79 group had statistical significance (P<0.01). With respect to gender, the difference was not statistically significant (P>0.05). Conclusion: The nonlongevity families have higher DNA damage frequency than longevity ones and the difference is statistical significant. Excluding the factors of age, sex, smoking and alcohol drinking, etc, the mean of DNA damage frequency in the two groups is still statistically significant ......
