三氧化二砷诱导子宫颈癌HeLa细胞凋亡的初步研究

摘要: 目的:研究三氧化二砷(As2O3)诱导子宫颈癌细胞系HeLa细胞凋亡,探讨As2O3对子宫颈癌的临床应用意义。方法:采用四甲基偶氮唑盐比色(MTT)法检测浓度分别为1.0、2.0、5.0、10.0 μmol/L的As2O3作用1、2、3、4 d后HeLa细胞的存活率,并进行形态学观察;采用细胞凋亡原位检测(TUNEL)法进行细胞凋亡的检测。结果:2.0 μmol/L As2O3处理HeLa细胞48 h后,可见细胞的存活率明显降低(P<0.05),处理96 h后,细胞的存活率进一步降低(P<0.01),经5.0 μmol/L As2O3处理的HeLa细胞24 h后就明显可见细胞的存活率降低;As2O3作用后,HeLa细胞具有明显的凋亡特征性改变;TUNEL法检测可发现HeLa细胞有DNA的断裂。结论:As2O3可以诱导子宫颈癌HeLa细胞凋亡,抑制细胞增殖,随着As2O3浓度增加,作用时间延长,效果越明显,其作用具有浓度和时间依赖性。关键词:三氧化二砷; 子宫颈癌; HeLa细胞;细胞凋亡
The primary study on programmed cell death of human uterin cervical cancer induced by arsenic trioxide

Abstract:Objective: To investigate the programmed cell death of human uterin cervical cancer induced by arsenic trioxide (As2O3) and research the arsenic trioxide significance of clinical application. Methods: Morphlogy investigation,MTT reduction assay and in situ cell death detection (TdTmediate Dutp nick labeling, TUNEL) were adopted. Results: The survival of HeLa cell line was inhibited by As2O3. And a typical apoptotic morphlogy also could be seen during the cells treated with As2O3. Moreover DNA fragmentation could be detected by TUNEL. Conclusion: As2O3 can induce programmed cell death of HeLa cells and inhibit the cellular proliferation. The effect shows time and concentration dependence. ......
