摘要:目的:探讨脓毒症大鼠肿瘤坏死因子a(TNFa)与肝细胞凋亡的关系以及对肝功能的影响。方法:大鼠随机分为正常对照组和实验组,实验组采用盲肠结扎穿孔术(CLP)致脓毒症大鼠模型,分别于术后30、60、120、180 min(CLP术后30、60、120、180 min组)处死动物,测定肝组织和血浆TNFa水平,并检测肝细胞凋亡的情况,同时测定丙氨酸氨基转移酶(ACL)和天冬氨酸氨基转移酶(AST)水平。正常对照组麻醉后不行CLP术,其余同实验组。 结果: 与正常对照组相比,CLP术后30、60、120、180 min组肝细胞凋亡数、血清TNFα、ALT水平均明显升高(P<0.01),有随着时间的延长增加的趋势;与正常对照组相比,CLP术后120、180 min组血清AST含量明显升高(P<0.01);CLP术后大鼠肝组织、TNFα表达强度随时间延长而有增强的趋势。结论: CLP后大鼠肝脏产生大量TNFα,与肝细胞凋亡有密切关系,肝功能受到损伤。关键词: 脓毒症; 肝细胞凋亡; 肿瘤坏死因子 α
The research of TNFα and hepatic apoptosis in sepsic rat

Abstract: Objective: To investigate the relation of tumor necrosing foctorα (TNFα) and hepatis apoptosis and the effect of liver function in sepsis. Methods: Wistar rats were subjected to sepsis induced by cecal ligation and puncture (CLP), and animals were randomly divided into normal control (n=6), septic group (n=24). Animal were sacrificed at 30, 60, 120, 180 min after CLP. Serum and liver tissure TNFα protein levels and liver apoptosis were measured at various intervals. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were also determined at the same time. Results: TNFα protein levels were significantly elevated compared to normal control in liver tissure and serum(P<0.01) , but there was no obviously change between 30 min and 60 min after CLP. In addition the number of liver apoptosis progressively increased. Meanwhile, serum ALT and AST in septic group were markedly increased comparing to normal control (P<0.01). Conclusion: It existed a close relation between TNF and liver apoptosis,and made liver function damage after CLP. ......
