实验性ARDS大鼠心脏Gq/11蛋白的动态变化

基金项目:新疆维吾尔自治区少数民族科技骨干人才特殊培养专项基金项目(200126,200413)摘要: 目的: 观察急性呼吸窘迫综合征(ARDS)时大鼠心脏Gq/11蛋白的动态变化,从分子水平进一步阐明ARDS并发多器官功能障碍的发病机制。方法:选择雄性Wistar大鼠40只, 采用尾静脉注射油酸(oleic acid,OA 0.2 ml/kg) 法复制大鼠ARDS模型,并将其分为对照组(C组)和油酸组(OA组),又根据不同时限将其分为30、60、90、120 min 4个亚组,检测血气、平均动脉血压(MAP)、心功(LVSP、左室±dp/dtmax)、血浆及心肌MDA含量和LDH、CK活性;采用免疫印迹法检测心脏Gq/11蛋白含量。结果: OA组PaO2、MAP随着时间延长呈进行性降低(P<0.05~0.01),LVSP、左室±dp/dtmax逐渐降低,OA 90、120 min组显著低于C组(P<0.01)。OA组除30 min外,血浆MDA含量、LDH活性均较C组明显升高(P<0.01),血浆CK活性从注射OA 30 min后开始增高(P<0.01)。OA各组心肌MDA含量、LDH活性较C组高,随时间延长呈升高趋势(P<0.05~0.01),心肌CK活性除OA 30 min组外均较C组高(P<0.01)。OA组除30 min外,Gq/11蛋白含量随时间延长较C组分别高(24.72±24.05)%、(40.61±22.41)%、(50.84±24.49)%。形态学观察: OA 30 min组心肌间质轻度水肿, OA 60 min组有少量炎性细胞浸润,部分心肌细胞溶解、坏死,OA 90、120 min组出现较多炎性细胞浸润。结论:Gq/11蛋白的高表达变化可能参与了ARDS过程并发心肌损害的病理性信号传导。

The dynamic alteration of Gq/11 protein concentration in the heart of rats
during acute respiratory distress syndrome

Abstract: Objective: The dynamic alteration of expression of Gq/11 protein in the heart of rats was studied during acute respiratory distress syndrome(ARDS) to explore the pathophysiological mechanism of ARDS induced multiple organ dysfunction syndrome (MODS) from molecular level. Methods: selecting 40 male Wistar rats, to establish an ARDS model by intravenous injection of oleic acid (OA 0.2 ml/Kg-1) though tail vein. These rats were randomly divided into experimental groups (OA group) and control group (C group). OA group include 4 subsections: OA 30, 60, 90, 20min. Blood gas indexes, cardiac function indexes (MAP, LVSP,±LV dp/dtmax), and enzyme indexes(MDA, LDH , CK, ACE) of plasma and heart were measured. The concentrations of Gq/11 in heart were measured by Western blot. Results: Compared with C group, PaO2 of OA groups obviously decreased (P<0.01). MAP of OA group reduced (P<0.05~0.01). With time passing, cardiac function indexes (LVSP, ±LV dp/dtmax) of OA groups gradually decreased. The indexes of OA 90 min, 120 min groups were lower those of C group (P<0.01). MDA content, LDH activity and CK activity of plasma and heart of OA groups (except OA30 min group) gradually increased (P<0.01). The concentration of Gq/11 in the heart of OA groups (except OA30 min group) respectively increased to (124.72±24.05)%, (140.61±22.41)%, (150.84±24.49)%(P<0.05~0.01). Morphologic observation: myocardium mesenchymal of OA 30 min has light edema; there are few of inflammatory cells and part of myocardial cells occur lysis and necrosis in the OA 60 min; there are more inflammatory cells in the OA90 min and OA120 min; the edema of mesenchymal is relieved and the amount of inflammatory cells decreased in the Pgroup. Conclusions: Up regulation of the expression of Gq/11 may play a role in heart injury during ARDS. ......
