人肺腺癌多药耐药细胞系的建立及其生物学性状的测定

摘要:目的:体外建立人肺腺癌细胞多药耐药细胞系。方法:以阿霉素(adriamycin, ADM)为诱导药,人肺腺癌细胞系(SPCA1)为诱导对象,逐步增加ADM药物浓度进行诱导,以一定浓度ADM培养46周后,光镜观察细胞形态,采用MTT法检测细胞耐药指数(RF),并同时对长春地辛、足叶乙甙、顺铂、5氟脲嘧啶、紫杉醇、ADM 6种药物进行交叉耐药检测。结果:SPCA1/ADM形态不规则,SPCA1/ADM对ADM的RF为11.3,同时对长春地辛、足叶乙甙、顺铂、5氟脲嘧啶、紫杉醇5种抗癌药物有不同程度的耐药性。结论:建立了相对稳定多药耐药细胞系SPCA1/ADM,可用于多药耐药性机制及逆转的研究。关键词:人肺腺癌细胞系; 多药耐药; 阿霉素; 诱导
