【摘要】 目的 研究中药党参、丹参、黄芪、生地和复方神肌再生冲剂对大鼠坐骨神经损伤后神经再生的影响。方法 将大鼠分为党参组、丹参组、黄芪组、生地组、复方冲剂组和对照组，每组大鼠18只。麻醉下暴露右侧坐骨神经，在坐骨结节远侧0.8cm 处造成坐骨神经挤压伤，根据分组，术后给予不同的中药喂养大鼠。术后不同时间，取紧邻挤压伤部位远侧神经干，应用原位杂交的方法和图像分析处理系统定量测定神经组织中神经生长因子mRNA(mRNANGF)的表达。结果 丹参组和黄芪组大鼠坐骨神经挤压伤后第2周起，坐骨神经组织中mRNANGF表达明显增加，至伤后第4周达到高峰。党参治疗坐骨神经挤压伤，治疗后8周，mRNANGF表达较对照组明显增加，差异有显著性。在复方冲剂组，大鼠坐骨神经挤压伤后8周内，神经组织中mRNANGF表达始终处于高水平。各组大鼠坐骨神经mRNANGF表达与对照组之间的差异具有显著性(P<0.05)。复方冲剂组大鼠坐骨神经mRNANGF表达与丹参和黄芪组之间的差异也具有显著性。结论 中药党参、丹参和黄芪可以促进挤压伤后大鼠坐骨神经mRNANGF表达；这种作用在组成复方冲剂后，表现得更为明显。

    Effects of Chinese herb on expression of mRNANGF after sciatic nerve injury in rats

    【Abstract】 Objective To decide which of Chinese herbs including Dangshen,Danshen,Huangqi and Shengdi has effect on improving the regeneration of peripheral nerve after nerve lesion and to study the mechanism of Chinese herb.Methods 108 SD rats(male,220～250g)were divided into six groups:Dangshen group,Danshen group,Huangqi group,Shengdi group,mixed group and control group.The rats were anaesthetized and the right sciatic nerves were crushed at the site 0.8cm distal to sciatic notch.After surgery the rats were fed with different Chinese herb according to different group.At various time after operation the rat were killed and the sciatic nerve stump just distal to the crush site were removed to evaluate the level of mRNANGF.mRNANGF was expressed by means of in situ hybridization and the level was determined by imaging analysis.Results In Danshen and Huangqi groups the levels of mRNANGF in sciatic nerve markedly increased at the second week after surgery.As compared with control group,the levels of mRNANGF in these two groups were greatly higher and the difference are statistically significant(P<0.05).In mixed group the mRNANGF are all in high level compared with control group.At the eighth week after crushed the expression of mRNANGF in Dangshen group was higher than that of control group.Conclusion Chinese herbs including Danshen,Huangqi,Dangshen and mixed herbs can increase the expression of mRNANGF of rat sciatic nerve and improve the regeneration of sciatic nerve after nerve crushed. ......
