热处理抗原冲击脐血细胞与肿瘤免疫效应的关系

摘要:目的 比较加热和不加热两种方法提取的肿瘤抗原冲击脐血有核细胞诱发的抗肿瘤免疫效应。方法收集对数生长期的肿瘤细胞SPCA1(人肺腺癌细胞),分为两组,一组42℃加热12 h,另一组不加热作对照组。两组细胞用超声粉碎机破碎后高速离心收集上清液,即制备成肿瘤抗原。结果 在体外,经加热处理的肿瘤抗原冲击脐血有核细胞的抗肿瘤效应明显优于未加热组,抗原浓度以5 μg/ml最为适宜。结论 肿瘤细胞经加热处理后可获得更强的抗肿瘤免疫效应。关键词:脐带血;热休克蛋白;肿瘤
Relation between heat treating antigen pulesd umbilical cord blood cells and tumor immune potency

Abstract:Objective Antitumor immune potency inducing tumor antigen pulsed umbilical cord blood cells was compared in two different ways. Methods Tumor cells SPCA1 was collected and separated into two groups. One was heat shock treatment for 12 hours at 42℃,and the other was control group. Supernatant of SPCA1 cells homogenization was tumor antigen.Results In vitro, heat shock treatment antigen to pulse umbilical cord blood with nucleus has much higher antitumor activity than control group, the suitable antigen concentration was 5 μg/ml. Conclusion Tumor antigen of heat shock treatment has high antitumor potency. ......
