摘要: 目的 建立脂质体凝胶剂中磷脂的定量测定方法。方法 将样品用硝酸、高氯酸消化,调pH值后,加入钼蓝法显色剂,在830 nm处比色。并进行了线性、精密度、稳定性和加样回收率等项考察。结果 该方法线性方程为: A = 36.109ρ–0.0148,在4 ~ 20 mg·L-1内,线性关系良好(r = 0.999 4),该方法精密度为:1.59%;显色后2 h比色结果稳定;加样回收率为:98.4% ~ 103.7%。结论 该方法适合于脂质体凝胶剂中磷脂含量的测定。

Determinating contents of phospholipids in liposomal gel with molybdenum blue method

Abstract: Objective To develop a method of determinating the contents of phospholipids in liposomal gel. Methods The sample was digested with nitric acid and perchloric acid, adjusted pH, added molybdenum blue reagent after adjusted pH,then colormetered at 830nm. In addition, the precision, stability, and the recovery of the method were also checked. Results The standard curve was A=36.109ρ-0.0148,with a good correlation in the concentration range of 4~20mg·L-1. The precision of the method was 1.59%(RSD), the absorption value was stable in 2h after color development. Conclusion This method is simple and is fit to determine of phospholipids contents in liposomal gel with good reproducibility.
