动力性肺动脉高压犬肺动脉PCNA及α┐平滑肌肌动蛋白的表达

关键词 :高血压,肺性;疾病模型,动物;血液动力学;增殖细胞核抗原;肌动蛋白类摘 要:目的 探讨平滑肌细胞增殖在动力性肺动脉高压形成中的作用. 方法 利用降主动脉—左下肺动脉分流建立的幼犬动力性肺动脉高压模型,取90d肺组织病理切片,应用鼠抗人增殖细胞核抗原(PCNA)抗体、α-平滑肌细胞肌动蛋白抗体(α-actin)、及Dako二抗免疫组化检测,图像分析仪对所得结果进行分析,然后计数高倍镜下平滑肌细胞总数及PC-NA细胞阳性数,计算细胞增殖指数. 结果 分流犬左下肺动脉中膜α-actin免疫组化染色呈强阳性表达,右下肺动脉弱阳性,而对照组均呈阴性;分流犬左下肺动脉PCNA阳性信号表达明显多于右肺动脉,而右肺动脉及左上肺动脉仅有少量表达;增殖指数左下肺动脉内膜为33%,中膜11%,外膜7%,右下肺动脉仅为少量. 结论 平滑肌细胞增殖参与实验犬动力性肺动脉高压形成过程并与肺动脉压力增高程度和肺血管重构改变一致.
