    摘 要:目的 建立一种无血清传代培养黑素细胞的方法，并观察抗角蛋白自身抗体(AK auto Ab)对无血清培养黑素细胞及角质形成细胞增殖的影响. 方法 应用角质形成细胞无血清培养基(KC-SFM)添加牛垂体浸出液(BPE)和TPA传代培养黑素细胞，应用KC-SFM添加EGF和BPE传代培养角质形成细胞，培养均至第3代时于培养基中加入不同浓度的AK auto Ab，用MTT法观察AK auto Ab对两种细胞增殖的影响. 结果 应用无血清培养基成功地传代培养了黑素细胞，MTT法结果显示AK auto Ab各浓度组对培养黑素细胞的增殖无明显影响，但对角质形成细胞的增殖有显著抑制作用并有浓度依赖性. 结论 建立了一种无血清培养黑素细胞的方法，AK auto Ab对黑素细胞的增殖无抑制作用而对角质形成细胞有特异性抑制作用.

    Abstract:AIM To observe the effect of anti-keratin autoan-tibody(AK auto Ab)on proliferation of melanocytes and keratinocytes cultured with serum-free medium.METHODS Human melanocytes were cultured with KC-SFM supple-mented with BPE and TPA while human keratinocytes were cultured with KC-SFM in the presence of EGF and BPE.Ef-fects of different titers of AK auto Ab in medium on prolifer-ation of melanocytes and keratinocytes were observed by means of MTT.RESULTS AK auto Ab had no effects on melanocytes but markedly inhibited the growth of target cells ......
