木黄酮对人垂体腺瘤细胞增殖的影响

关键词: 木黄酮;垂体腺瘤细胞;体外培养摘 要:目的 观察木黄酮(genistein,GST)对培养的人垂体腺瘤细胞增殖的影响. 方法 将GST作用于体外培养的人垂体腺瘤细胞,测定MTT值及3 H-TdR参入量,并用流式细胞仪测定细胞周期. 结果 不同浓度的GST(10-7 ,10-6 和10-5 mol·L-1 )明显抑制垂体腺瘤细胞的增殖,MTT的A490nm 值变化率分别为(91.2±4.9)%,(78.8±5.3)%和(65.8±5.6)%,(P<0.01),cpm值的变化率分别为(88.6±3.6)%,(75.5±5.7)%和(58.9±4.9)%,(P<0.01),并存在着剂量效应.10-5 mol·L-1 的GST可使G1期细胞比例从对照组的68.2%上升为91.0%(P<0.01).木黄酮对垂体腺瘤细胞的作用与雌激素受体无关. 结论 GST在体外能明显抑制培养的人垂体腺瘤细胞的增殖.
